America's Air Force Seeks Commercial Technologies For 'Space Domain Awareness' (spacenews.com) 27

America's Air Force is seeking proposals for technologies "for operations far beyond geosynchronous Earth orbit, near the moon's orbit," reports SpaceNews: Specific items the Air Force wants: payloads for providing space domain awareness from the lunar surface, lightweight sensors for space-based space domain awareness, and methodologies for orbit determination and catalog maintenance in cislunar space. The Air Force also is interested in concepts for providing position, navigation and timing solutions for cislunar space operations; visualization of cislunar orbits; and terrestrial-based concepts for achieving space domain awareness of cislunar space.

The inclusion of cislunar space capabilities in the Small Business Innovation Research program was unexpected, said Shawn Usman, an astrophysicist and founder of the space consulting firm Rhea Space Activity. The industry sees this as a sign that the Air Force, and the future Space Force, are responding to advances made by China, Usman told SpaceNews. "This is definitely a pretty big turning point for the U.S. Air Force, the U.S. 'new space' industry, our near-peer competitors, and for the upcoming Space Force."

America Considers Declassifying Military Information on US, Chinese, and Russian Space Programs (defensenews.com) 55

"Declassifying some of what is currently held in secure vaults would be a good idea," Air Force Secretary Barbara Barrett said during a panel at the Reagan National Defense Forum. "You would have to be careful about what we declassify, but there is much more classified than what needs to be."

Fellow panelist Rep. Mike Rogers, R-Ala., said he met with the secretary earlier in the week to discuss that very issue, calling the information on space programs "overwhelmingly classified." For Rogers, that overclassification is one of the reasons it's been so difficult for him and others to build support both in the public and with other members of Congress for a Space Force, a sixth branch of the military under the Air Force uniquely focused on space as a war-fighting domain.... "I don't think that can happen until we see significant declassification of what we're doing in space and what China and Russia are doing, and how space is in their day-to-day lives."

Barrett also argued that America's way of life "is more dependent on space than any other nation's. And our capability in space was predominantly built at a time when we thought space was a benign environment."

Physicists Use Bubbling Quantum Vacuum To Hopscotch Heat Across Empty Space (livescience.com) 23

That's the sensation of billions of atoms banging together. Tiny vibrations carry thermal energy from the water to the mug and then into your skin as one molecule knocks into the next, sending it careening into a third -- and so on down the line.

Heat can also cross space as waves of radiation, but without radiation, it needs stuff to pass through -- molecules to bang into other molecules. Vacuums have no 'stuff' in them, so they tend to trap heat. In Earth's orbit, for example, one of the biggest engineering challenges is figuring out how to cool down a rocket ship.

But now, researchers have shown that, on microscopic scales, this isn't really true. In a new paper published Dec. 11 in the journal Nature, physicists showed that little vibrations of heat can cross hundreds of nanometers of empty space. Their experiment exploited an uncanny feature of the quantum vacuum: It isn't really empty at all.

Astronomers Find 19 More Galaxies Missing Their Dark Matter (astronomy.com) 90

A reader shares a report from Astronomy.com: Astronomers have discovered 19 more galaxies missing their dark matter. Instead of dark matter, these strange galaxies are mainly filled with regular matter, like the protons, neutrons, and electrons that make up everything we're familiar with. The new find, published November 26 in Nature Astronomy, bolsters the controversial recent discovery of two other galaxies without dark matter. The mysterious substance accounts for most matter in the universe, and it's thought to be the primary component of all galaxies -- as well as the main driver of galaxy formation in the first place. So, finding so many galaxies without the exotic matter suggests astronomers are missing something major about how galaxies form and evolve.

"This result is very hard to explain using the standard galaxy formation model," said lead author Qi Guo of the Chinese Academy of Science in a press release, "and thus encourages people to revisit the nature of dark matter." The latest batch of galaxies missing dark matter was discovered when Guo and her team explored the nature of 324 dwarf galaxies using data from the Arecibo Observatory in Puerto Rico and the Sloan Digital Sky Survey. With this data, they followed in the footsteps of Rubin and Ford, studying how fast hydrogen gas rotates around each galaxy. They also calculated how much normal matter -- in the form of both gas and stars -- they contained. After crunching the numbers, Guo and her colleagues determined that, of the 324 dwarf galaxies they investigated, 19 of them contain enough visible matter to solely explain the motions of the galaxies' hydrogen. In other words, a lot of dark matter seems to be missing from these galaxies.

Tony Brooker, Pioneer of Computer Programming, Dies At 94 (nytimes.com) 26

Cade Metz from The New York Times pays tribute to Tony Brooker, the mathematician and computer scientist who designed the programming language for the world's first commercial computer. Brooker died on Nov. 20 at the age of 94. From the report: Mr. Brooker had been immersed in early computer research at the University of Cambridge when one day, on his way home from a mountain-climbing trip in North Wales, he stopped at the University of Manchester to tour its computer lab, which was among the first of its kind. Dropping in unannounced, he introduced himself to Alan Turing, a founding father of the computer age, who at the time was the lab's deputy director. When Mr. Brooker described his own research at the University of Cambridge, he later recalled, Mr. Turing said, "Well, we can always employ someone like you." Soon they were colleagues.

Mr. Brooker joined the Manchester lab in October 1951, just after it installed a new machine called the Ferranti Mark 1. His job, he told the British Library in an interview in 2010, was to make the Mark 1 "usable." Mr. Turing had written a user's manual, but it was far from intuitive. To program the machine, engineers had to write in binary code -- patterns made up of 0s and 1s -- and they had to write them backward, from right to left, because this was the way the hardware read them. It was "extremely neat and very clever but pretty meaningless and very unfriendly," Mr. Brooker said. In the months that followed, Mr. Brooker wrote a language he called Autocode, based on ordinary numbers and letters. It allowed anyone to program the machine -- not just the limited group of trained engineers who understood the hardware. This marked the beginning of what were later called "high-level" programming languages -- languages that provide increasingly simple and intuitive ways of giving commands to computers, from the IBM mainframes of the 1960s to the PCs of the 1980s to the iPhones of today.

A Space Probe Has Mapped the Winds Above Mars for the First Time (technologyreview.com) 5

Scientists have mapped out Mars's upper atmosphere wind patterns for the first time. The findings, published Thursday in Science, reinforce our understanding of the Martian climate as equal parts stable and unpredictable. From a report: The investigation uses data collected by NASA's Mars Atmosphere and Volatile Evolution (MAVEN) mission, which has been orbiting Mars since 2013. MAVEN has helped teach us how Mars lost its thick atmosphere billions of years ago, but it was never designed to investigate winds. Instead, the team behind the new study had a clever idea: have MAVEN rapidly swing its normally stationary Natural Gas and Ion Mass Spectrometer (NGIMS) back and forth like a windshield wiper. This swinging effect meant that NGIMS, usually used to study atmospheric chemistry, was able to offset the orbiter's own movements and measure the winds as if it were standing still. What did they find? Overall circulation patterns in Mars's upper atmosphere proved predictably stable season-to-season. But the team also found extreme variability within local pockets of the atmosphere, and so far there's no good explanation for what's causing this.

Mathematicians regard the Collatz conjecture as a quagmire and warn each other to stay away. But now Terence Tao has made more progress than anyone in decades. From a report: It's a siren song, they say: Fall under its trance and you may never do meaningful work again. The Collatz conjecture is quite possibly the simplest unsolved problem in mathematics -- which is exactly what makes it so treacherously alluring. "This is a really dangerous problem. People become obsessed with it and it really is impossible," said Jeffrey Lagarias, a mathematician at the University of Michigan and an expert on the Collatz conjecture. Earlier this year one of the top mathematicians in the world dared to confront the problem -- and came away with one of the most significant results on the Collatz conjecture in decades. On September 8, Terence Tao posted a proof showing that -- at the very least -- the Collatz conjecture is "almost" true for "almost" all numbers. While Tao's result is not a full proof of the conjecture, it is a major advance on a problem that doesn't give up its secrets easily. "I wasn't expecting to solve this problem completely," said Tao, a mathematician at the University of California, Los Angeles. "But what I did was more than I expected."

Lothar Collatz likely posed the eponymous conjecture in the 1930s. The problem sounds like a party trick. Pick a number, any number. If it's odd, multiply it by 3 and add 1. If it's even, divide it by 2. Now you have a new number. Apply the same rules to the new number. The conjecture is about what happens as you keep repeating the process. Intuition might suggest that the number you start with affects the number you end up with. Maybe some numbers eventually spiral all the way down to 1. Maybe others go marching off to infinity. But Collatz predicted that's not the case. He conjectured that if you start with a positive whole number and run this process long enough, all starting values will lead to 1. And once you hit 1, the rules of the Collatz conjecture confine you to a loop: 1, 4, 2, 1, 4, 2, 1, on and on forever.

Cigna Uses AI To Check If Patients Are Taking Their Medications (wsj.com) 32

An anonymous reader quotes a report from The Wall Street Journal: Cigna plans to expand a system that uses artificial intelligence to identify gaps in treatment of chronic diseases, such as patients skipping their medications, and deliver personalized recommendations for specific patients. The product, called Health Connect 360, integrates data from a combination of sources and analytical tools, some developed at Cigna and others brought in as part of its $54 billion acquisition of pharmacy-benefit manager Express Scripts Holding Co., completed late last year. Express Scripts, which began developing the service two years ago, rolled out portions of it to some customers this year.

Health Connect 360 was developed for treatment of chronic diseases, including diabetes and heart disease, as well as for pain management. The system aggregates medical, pharmacy, lab and biometric data -- such as information from glucometers, which measure blood-sugar levels -- into a dashboard that is accessible through an online interface. The dashboard will be visible to the service's customers and to Express Scripts case managers and nurses with access rights. The system can also feed information to electronic-medical record systems for physicians. Cigna is already using AI to predict whether patients might abuse or overdose on prescription opioids. Another Cigna tool, One Guide, provides personalized help to health-insurance holders on their benefit plans, appointments and health coaching. The new Health Connect 360 system combines algorithms that analyze data such as clinical and pharmacy information with predictive models to generate recommendations and ways to best engage a patient, whether through an app or in person.

New Zealand Orders 1,300 Square Feet of Skin From US To Help Burned Volcano Victims (people.com) 65

schwit1 shares a report from People: Doctors in New Zealand are currently awaiting nearly 1,300 square feet of skin from the United States in order to treat the dozens of victims who suffered severe burns when a volcano erupted on White Island Monday afternoon. Dr. Peter Watson, chief medical officer of Counties Manukau Health, said at a press conference Wednesday that there are 29 patients being treated in intensive care and burn units at four different hospitals throughout New Zealand. Twenty-four of the burn patients remain in critical condition.

"We currently have supplies but are urgently sourcing additional supplies to meet the demand for dressing and temporary skin grafts," Watson said. "We anticipate we will require an additional 1.2 million square centimeters [1,292 square feet] of skin for the ongoing needs of the patients. These supplies are coming from the United States and the order has been placed." Watson said the nature of the victims' injuries had been made "complicated" by the gases and chemicals in the eruption, thus making "more rapid" surgical treatment necessary, as opposed to if they'd suffered thermal-only burns.
CNN reports that the skin grafts are coming from people who are registered to donate skin after their deaths, and typically are taken from the donors' backs or the backs of their legs.

There were a total of 47 travelers on the island when the volcano erupted Monday just after 2 p.m. Six people were killed.

Crows Could Be the Smartest Animal Other Than Primates (bbc.com) 130

In a piece for the BBC, Chris Baraniuk writes about how the intelligence of New Caledonian crows may be far more advanced than we ever thought possible. An anonymous reader shares an excerpt from the report: Intelligence is rooted in the brain. Clever primates -- including humans -- have a particular structure in their brains called the neocortex. It is thought that this helps to make advanced cognition possible. Corvids, notably, do not have this structure. [New Caledonian crows belong to the corvid family of birds -- as do jackdaws, rooks, jays, magpies and ravens.] They have instead evolved densely packed clusters of neurons that afford them similar mental prowess. The specific kind of brain they have doesn't really matter -- corvids and primates share some of the same basic capabilities in terms of problem-solving and plasticity, or being able to adapt and change in the face of new information and experiences. This is an example of convergent evolution, where completely different evolutionary histories have led to the same feature or behavior. It's easy for humans to see why the things corvids can do are useful. From identifying people who have previously posed a threat to them or others in their group to using gestures for communication -- we too rely on abilities like these.

[Christian Rutz at the University of St Andrews] is unequivocal. Some birds, like the New Caledonian crows he studies -- can do remarkable things. In a paper published earlier this year, he and his co-authors described how New Caledonians seek out a specific type of plant stem from which to make their hooked tools. Experiments showed that crows found the stems they desired even when they had been disguised with leaves from a different plant species. This suggested that the birds were selecting a kind of material for their tools that they knew was just right for the job. You wouldn't use a spanner to hammer in a nail, would you? Ranking the intelligence of animals seems an increasingly pointless exercise when one considers the really important thing: how well that animal is adapted to its niche. In the wild, New Caledonians use their tools to scoop insects out of holes, for example in tree trunks. Footage of this behavior has been caught on camera.

You might think that some animals are smarter than others -- with humans at the top of the proverbial tree. Certainly, humans do rely excessively on intelligence to get by. But that doesn't mean we're the best at every mental task. Chimps, notes Dakota McCoy at Harvard University, have been shown to possess better short-term memories than humans. This might help them to memorize where food is located in the forest canopy, for example. Ranking the intelligence of animals seems an increasingly pointless exercise when one considers the really important thing: how well that animal is adapted to its niche. Intelligence is, first and foremost, a means towards specialization.
"New Caledonian crows, like us and other clever animals, have moods and memories. Strategies and expectations. They seem remarkably able to engage with complexity," writes Baraniuk in closing. "Evolution made this possible. But cognition, like life itself, serves more than just a need. Animal intelligence allows all sorts of fascinating phenomena to arise. [...] Nature provided the notes, but animal brains make the music. The mind, as they say, is the only limit."

Rude Paper Reviews Are Pervasive and Sometimes Harmful, Study Finds (sciencemag.org) 135

sciencehabit writes: There's a running joke in academia about Reviewer 2. That's the reviewer that doesn't bother to read the manuscript a journal has sent out for evaluation for possible publication, offers condescending or outright offensive comments, and -- of course -- urges the irrelevant citation of their own work. Such unprofessional conduct is so pervasive there's even a whole Facebook group, more than 25,000 members strong, named "Reviewer 2 Must Be Stopped!" But it is no laughing matter, concludes a new study that finds boorish reviewer comments can have serious negative impacts, especially on authors belonging to marginalized groups.

The study surveyed 1106 scientists from 46 countries and 14 disciplines. More than half of the respondents -- who were promised anonymity -- reported receiving at least one "unprofessional" review, and a majority of those said they had received multiple problematic comments. Those comments tended to personally target a scientist, lack constructive criticism, or were just unnecessarily harsh or cruel, the authors report. For example, one author received a review that stated: "The phrases I have so far avoided using in this review are 'lipstick on a pig' and 'bullshit baffles brains.'" Another reported receiving this missive: "The author's last name sounds Spanish. I didn't read the manuscript because I'm sure it's full of bad English."

Larry Page Is Quietly Funding Efforts To Develop a Universal Flu Vaccine (theverge.com) 57

An anonymous reader quotes a report from The Verge: Google co-founder Larry Page is funneling money from his charitable foundation to a private flu-fighting initiative run as a for-profit company, TechCrunch reported. The program offers free flu shots to children in Oakland, California-area schools. Page also has a second company funding efforts to create a universal flu vaccine, according to the report. The free flu shots are offered through a group called Shoo The Flu, which started funding flu shots for both elementary and kindergarten through eighth grade schools in 2014. Shoo the Flu reimburses the Alameda County Public Health Department and school districts for the cost of the program.

The second company, Flu Lab, provides funding for the Bill & Melinda Gates Foundation's Grand Challenge for Universal Influenza Vaccine Development. Page and his family were initially contributing funding to the challenge directly, but they were replaced by Flu Lab. Flu Lab also supports the nonprofit Sabin Vaccine Institute, which works to expand vaccine access. The funding established the Sabin-Aspen Vaccine Science & Policy Group, which met in 2018 to discuss efforts to develop a universal flu vaccine. Shoo the Flu's director is also the CEO of Flu Lab, and TechCrunch reported that Shoo the Flu will soon move under the umbrella of Flu Lab.
The report notes that both Shoo the Flu and Flu Lab are private, for-profit companies and therefore not required to file public tax returns.

"However, the funding for Shoo the Flu comes from Page's charitable foundation, the Carl Victor Page Memorial Foundation, which does have to file public returns," reports The Verge. "That makes the money flowing into the company public; Page's foundation gave Shoo the Flu over $4.1 million between 2015 and 2017, TechCrunch reported. What the organization did with that money is not as readily available."

Low-Risk Ultrasound Procedure Destroys 80 Percent of Prostate Cancers In One-Year Study (slashgear.com) 78

An anonymous reader quotes a report from SlashGear: A new treatment shows promise for revolutionizing prostate cancer treatment, offering a minimally-invasive and relatively low-risk alternative to traditional surgeries and radiotherapies. Called TULSA, this method uses sound waves to eliminate the diseased tissue in the prostate, leaving the rest of the healthy tissues behind. According to the researchers, patients treated with this method experience "minimal side effects." The transurethral ultrasound ablation (TULSA) method uses an MRI to guide the procedure, which involves inserting a rod through the urethra into the prostate, where it uses heat via sound waves to destroy the cancerous tissues. Unlike the surgery typically used to treat this condition, TULSA is minimally invasive and can be performed as an outpatient procedure.

Using guided and controlled sound waves, doctors are able to preserve the nerves near the prostate while eliminating the diseased tissues using a total of 10 elements located on the insertable rod. A software algorithm is part of the system -- it controls the strength, direction, and shape of the ultrasound beam, though doctors watch carefully using the MRI in real-time. A new study involving 115 men found that the average treatment time for this procedure is a bit less than an hour. The researchers found that 80-percent of patients experienced elimination of "clinically significant" cancer and that 72 of the men had no signs of cancer after the first year. As well, incontinence was a very rare side effect of the procedure, which also had low instances of impotence.

Chronic Pain is an Impossible Problem (theatlantic.com) 180

A "safe" alternative to opioid painkillers turns out to be not so safe. From a report: Gabapentin was supposed to be the answer. Chronic pain afflicts about a fifth of American adults, and for years, doctors thought it could be treated with prescription painkillers like Oxycontin. But as the drugs began killing the equivalent of three planeloads of Americans every week, opioid prescriptions fell off precipitously. Many doctors embraced gabapentin, an anticonvulsant drug traditionally used to prevent seizures, as a way to treat neuropathic pain while avoiding triggering life-threatening addiction. From 2012 to 2016, prescriptions for gabapentin increased 64 percent. It's now the 10th-most-commonly-prescribed medication in the United States. Baclofen, a muscle relaxant, has become another popular opioid replacement. Though gabapentin and baclofen can cause a boozelike "high" for some people, they're far less addictive and less likely to be fatal when taken in large quantities than opioids are.

But now their own pitfalls are becoming clear. Though gabapentin and baclofen are much safer alternatives to opioids, recent research suggests that they're not as safe as some doctors might have hoped, especially in combination with other sedating medications. The findings are a frustrating turn that suggests there's still no silver bullet for chronic pain. By examining the National Poison Data System, which collects reports of poisonings around the United States, Kimberly Reynolds, a researcher at the University of Pittsburgh, and her co-authors recently found that people are increasingly using both gabapentin and baclofen to either get high or attempt suicide. From 2013 to 2017, people tried to commit suicide using gabapentin nearly 42,000 times, and thousands more abused or misused the substance. In most cases -- nearly 70 percent -- the poisoned individuals took a combination of gabapentin and other drugs. Meanwhile, the majority of the poison cases involving baclofen were suicide attempts. "It's rare, but overdose from large quantities of gabapentin or baclofen can be fatal," Reynolds told me via email.

Gamma-Ray Laser Moves a Step Closer To Reality (phys.org) 85

A physicist at the University of California, Riverside, has performed calculations showing hollow spherical bubbles filled with a gas of positronium atoms are stable in liquid helium. The calculations take scientists a step closer to realizing a gamma-ray laser, which may have applications in medical imaging, spacecraft propulsion, and cancer treatment. Phys.Org reports: Extremely short-lived and only briefly stable, positronium is a hydrogen-like atom and a mixture of matter and antimatter -- specifically, bound states of electrons and their antiparticles called positrons. To create a gamma-ray laser beam, positronium needs to be in a state called a Bose-Einstein condensate -- a collection of positronium atoms in the same quantum state, allowing for more interactions and gamma radiation. Such a condensate is the key ingredient of a gamma-ray laser.

"My calculations show that a bubble in liquid helium containing a million atoms of positronium would have a number density six times that of ordinary air and would exist as a matter-antimatter Bose-Einstein condensate," said Allen Mills, a professor in the Department of Physics and Astronomy and sole author of the study that appears today in Physical Review A. [...] Mills, who directs the Positron Laboratory at UC Riverside, said the lab is configuring an antimatter beam in a quest to produce the exotic bubbles in liquid helium that Mills' calculations predict. Such bubbles could serve as a source of positronium Bose-Einstein condensates.

Greenland's Ice Sheet Melting Seven Times Faster Than In 1990s (theguardian.com) 282

An anonymous reader quotes a report from The Guardian: Greenland's ice sheet is melting much faster than previously thought, threatening hundreds of millions of people with inundation and bringing some of the irreversible impacts of the climate emergency much closer. Ice is being lost from Greenland seven times faster than it was in the 1990s, and the scale and speed of ice loss is much higher than was predicted in the comprehensive studies of global climate science by the Intergovernmental Panel on Climate Change, according to data. That means sea level rises are likely to reach 67cm by 2100, about 7cm more than the IPCC's main prediction. Such a rate of rise will put 400 million people at risk of flooding every year, instead of the 360 million predicted by the IPCC, by the end of the century.

Greenland has lost 3.8 trillion tons of ice since 1992, and the rate of ice loss has risen from 33 billion tons a year in the 1990s to 254 billion tons a year in the past decade. Greenland's ice contributes directly to sea level rises as it melts because it rests on a large land mass, unlike the floating sea ice that makes up much of the rest of the Arctic ice cap. About half of the ice loss from Greenland was from melting driven by air surface temperatures, which have risen much faster in the Arctic than the global average, and the rest was from the speeding up of the flow of ice into the sea from glaciers, driven by the warming ocean. The scale and speed of the ice loss surprised the team of 96 polar scientists behind the findings, published on Tuesday in the journal Nature. The Ice Sheet Mass Balance Inter-comparison Exercise comprised 26 separate surveys of Greenland from 1992 to 2018, with data from 11 different satellites and comparisons of volume, flow and gravity compiled by experts from the UK, Nasa in the US, and the European Space Agency.

Using a divide-and-conquer approach that leverages the power of compressed sensing, computer scientists from Rice University and Amazon have shown they can slash the amount of time and computational resources it takes to train computers for product search and similar "extreme classification problems" like speech translation and answering general questions. Tech Xplore reports: In tests on an Amazon search dataset that included some 70 million queries and more than 49 million products, Shrivastava, Medini and colleagues showed their approach of using "merged-average classifiers via hashing," (MACH) required a fraction of the training resources of some state-of-the-art commercial systems. "Our training times are about 7-10 times faster, and our memory footprints are 2-4 times smaller than the best baseline performances of previously reported large-scale, distributed deep-learning systems," said Shrivastava, an assistant professor of computer science at Rice. Medini, a Ph.D. student at Rice, said product search is challenging, in part, because of the sheer number of products. "There are about 1 million English words, for example, but there are easily more than 100 million products online."

MACH takes a very different approach [than current training algorithms]. Shrivastava describes it with a thought experiment randomly dividing the 100 million products into three classes, which take the form of buckets. "I'm mixing, let's say, iPhones with chargers and T-shirts all in the same bucket," he said. "It's a drastic reduction from 100 million to three." In the thought experiment, the 100 million products are randomly sorted into three buckets in two different worlds, which means that products can wind up in different buckets in each world. A classifier is trained to assign searches to the buckets rather than the products inside them, meaning the classifier only needs to map a search to one of three classes of product. [...] In their experiments with Amazon's training database, Shrivastava, Medini and colleagues randomly divided the 49 million products into 10,000 classes, or buckets, and repeated the process 32 times. That reduced the number of parameters in the model from around 100 billion to 6.4 billion. And training the model took less time and less memory than some of the best reported training times on models with comparable parameters, including Google's Sparsely-Gated Mixture-of-Experts (MoE) model, Medini said. He said MACH's most significant feature is that it requires no communication between parallel processors. In the thought experiment, that is what's represented by the separate, independent worlds.
The research will be presented this week at the 2019 Conference on Neural Information Processing Systems (NeurIPS 2019) in Vancouver.

Scientists Find a Weak Spot In Some Superbugs' Defenses (wired.com) 27

Researchers have found a new way to attack some of the bacteria behind treatment-resistant infections. An anonymous reader shares a report from Wired: In 2004, a 64-year-old woman in Indiana had a catheter put in to help with dialysis. Soon after the procedure, she came to a local hospital with low blood pressure and what turned out to be a dangerous antibiotic-resistant infection from a bacteria called Enterococcus faecalis. [...] After the patient in Indiana returned to the hospital, doctors sampled her blood and tested various antibiotics to see what might cure her infection. The strain she was infected with was already resistant to the antibiotic vancomycin, which was traditionally considered the treatment of last resort. But the bacteria that were making her sick were susceptible to a powerful new drug, approved by the FDA just a year before, called daptomycin. With a prescription for daptomycin, the patient improved enough to go home.

But two weeks later, the woman was back in the hospital again, this time with a high fever. Nothing her medical team tried worked, and the woman died. A study out today in the Proceedings of the National Academy of Sciences, though, offers new hope -- along with clues about how drug developers might fight back against this foe. VRE bacteria reproduce by pinching in at the center and dividing into two separate cells. Daptomycin fights VRE by binding to its cell membrane right at that center point, which disrupts its ability to divide, among other things. After the patient in Indiana died, doctors compared a sample of her blood with one they'd taken weeks earlier, when she first came to the hospital. They discovered that the daptomycin-resistant strain had a new mechanism that reorganized the cell. Daptomycin could no longer attach and halt the bacteria's cell division.
"[The researchers] were puzzled that the cells somehow knew when to organize their membranes to resist the daptomycin," reports Wired. "[Researcher Ayesha Khan] noticed these drug-resistant strains had a lot of the protein LiaX both on the cell membranes and outside the cell, so she zeroed in on it. LiaX, the research team found, is an alarm system. The protein binds to daptomycin, sending a signal back telling the cell that it's time to reorganize. This same mechanism also helps VRE ward off the human immune system, they found, which might contribute to its deadly nature."

"We knew prior to this study that LiaX likely has a role in daptomycin resistance, and this work goes a long way toward explaining what that role is," Kelli Palmer, a biologist who studies antibiotic resistance at the University of Texas at Dallas, said. "It is critical that we understand how daptomycin resistance works at a molecular level, so that we can design strategies to reverse it."
