Physical Force Alone Spurs Gene Expressions, Study Reveals (phys.org) 21

An anonymous reader quotes a report from Phys.Org: Cells will ramp up gene expression in response to physical forces alone, a new study finds. Gene activation, the first step of protein production, starts less than one millisecond after a cell is stretched -- hundreds of times faster than chemical signals can travel, the researchers report. The scientists tested forces that are biologically relevant -- equivalent to those exerted on human cells by breathing, exercising or vocalizing. They report their findings in the journal Science Advances.

In the new work, the researchers observed that special DNA-associated proteins called histones played a central role in whether gene expression increased in response to forces that stretched the cell. Histones regulate DNA, winding it up to package it in the nucleus of the cell. One class of histones, known as Histone H3, appear to prevent force-responsive gene expression when methylated at an amino acid known as lysine 9. Methylation involves adding a molecular tag known as a methyl group to a molecule. The scientists observed that H3K9 methylation was highest at the periphery of the nucleus and largely absent from the interior, making the genes in the interior more responsive to stretching. The researchers found they could suppress or boost force-responsive gene expression by increasing or decreasing H3K9 histone methylation. The scientists also tested whether the frequency of an applied force influenced gene expression. They found that cells were most responsive to forces with frequencies up to about 10-20 hertz.

SpaceX Loses Its Third Starship Prototype During a Cryogenic Test (arstechnica.com) 58

For a third time, SpaceX lost one of its Starship prototype spacecrafts during a pressure test at the company's test site in Boca Chica, Texas. Ars Technica reports: This week, SpaceX workers in South Texas loaded the third full-scale Starship prototype -- SN3 -- onto a test stand at the company's Boca Chica launch site. On Wednesday night, they pressure-tested the vehicle at ambient temperature with nitrogen, and SN3 performed fine. On Thursday night SpaceX began cryo-testing the vehicle, which means it was loaded again with nitrogen, but this time it was chilled to flight-like temperatures and put under flight-like pressures. Unfortunately, a little after 2am local time, SN3 failed and began to collapse on top of itself. It appeared as if the vehicle may have lost pressurization and become top-heavy. Multiple sources indicated that had these preliminary tests succeeded, SN3 would have attempted a 150-meter flight test as early as next Tuesday. SpaceX founder Elon Musk said on Twitter: "We will see what data review says in the morning, but this may have been a test configuration mistake." A testing issue would be good in the sense that it means the vehicle itself performed well, and the problem can be more easily addressed.

Potential Vaccine Generates Enough Antibodies To Fight Off Virus (independent.co.uk) 120

Slashdot readers schwit1 and Futurepower(R) are sharing news about a potential coronavirus vaccine that has been found to produce antibodies capable of fighting off Covid-19. The Independent reports: The vaccine, which was tested on mice by researchers at the University of Pittsburgh School of Medicine, generated the antibodies in quantities thought to be enough to "neutralize" the virus within two weeks of injection. The study's authors are now set to apply to the U.S. Food and Drug Administration for investigational new drug approval ahead of phase one human clinical trials planned to start in the next few months. [T]he Pittsburgh research is the first study on a Covid-19 vaccine candidate to be published after review from fellow scientists at outside institutions. The scientists were able to act quickly because they had already laid the groundwork during earlier epidemics of coronaviruses: Sars in 2003 and Mers in 2014. What's also neat about this potential vaccine is that it can sit at room temperature until it is needed and be scaled up to produce the protein on an industrial scale.

The fingertip-sized patch of 400 tiny microneedles "inject the spike protein pieces into the skin, where the immune reaction is strongest," the report says. "The patch is stuck on like a plaster and the needles -- which are made entirely of sugar and the protein pieces -- simply dissolve into the skin." While long-term testing is still required, "the mice who were given the Pittsburgh researchers' Mers vaccine candidate developed enough antibodies to neutralize the virus for at least a year," reports The Independent. "The antibody levels of the rodents vaccinated against Covid-19 'seem to be following the same trend,' according to the researchers."

Trump: CDC Recommends Cloth Face Covering To Protect Against Coronavirus 145

President Trump says the CDC now recommends using a cloth face covering to protect against coronavirus, but said he does not plan to do so himself. CNBC reports: Trump stressed that the recommendations were merely voluntary, not required. "I don't think I'm going to be doing it" he said as he announced the new guidance. The CDC's website explained that the recommendations were updated following new studies that some infected people can transmit the coronavirus even without displaying symptoms of the disease.
"In light of this new evidence, CDC recommends wearing cloth face coverings in public settings where other social distancing measures are difficult to maintain," such as in grocery stores or pharmacies, "especially in areas of significant community-based transmission," the CDC says.

Modern Meteorology Was Born 60 Years Ago Today (arstechnica.com) 19

"Sixty years ago on this date, April 1, a Thor-Able rocket launched a small satellite weighing 122.5kg into an orbit about 650km above the Earth's surface," writes Ars Technica's Eric Berger. "Effectively, this launch from Florida's Cape Canaveral Air Force Station marked the beginning of the era of modern weather forecasting." From the report: Designed by the Radio Corporation of America and put into space by NASA, the Television InfraRed Observation Satellite, or TIROS-1, was the nation's first weather satellite. During its 78 days of operation, TIROS-1 successfully monitored Earth's cloud cover and weather patterns from space. This was a potent moment for the field of meteorology. For the first time, scientists were able to combine space-based observations with physical models of the atmosphere that were just beginning to be run on supercomputers.

After World War II, mathematician John von Neumann led development of a computer to crunch through a set of equations put together by Jule Charney and other scientists. By the mid-1950s, Charney's group began to produce numerical forecasts on a regular basis. All of a sudden, meteorologists had two incredibly useful tools at their hands. Of course, it would take time for more powerful computers to produce higher-resolution forecasts, and the sensor technology launched on satellites would require decades to improve to the point where spacecraft could collect data for temperature, moisture, and other environmental variables at various levels in the atmosphere. But by around 1980, the tools of satellite observations and numerical models that could process that data started to mature. Scientists had global satellite coverage, 24 hours a day, and forecasts began to improve dramatically. Today, the fifth day of a five-day forecast on the app on your phone is about as accurate as the next day's forecast was in 1980.

New Blood Test Can Detect 50 Types of Cancer (theguardian.com) 24

A new blood test that can detect more than 50 types of cancer has been revealed by researchers in the latest study to offer hope for early detection. From a report: The test is based on DNA that is shed by tumours and found circulating in the blood. More specifically, it focuses on chemical changes to this DNA, known as methylation patterns. Researchers say the test can not only tell whether someone has cancer, but can also shed light on the type of cancer they have. Dr Geoffrey Oxnard of Boston's Dana-Farber Cancer Institute, part of Harvard Medical School, said the test was now being explored in clinical trials. "You need to use a test like this in an independent group at risk of cancer to actually show that you can find the cancers, and figure out what to do about it when you find them," he said.

Writing in the journal Annals of Oncology, the team reveal how the test was developed using a machine learning algorithm -- a type of artificial intelligence. Such systems pick up on patterns within data and as a result learn to classify it. The team initially fed the system with data on methylation patterns in DNA from within blood samples taken from more than 2,800 patients, before further training it with data from 3,052 participants, 1,531 of whom had cancer and 1,521 of whom did not.

Freshly Exhumed shares a report from CBC.ca: "[Autoclaving] is like a pressure cooker -- basically you enclose items into a chamber, you lock down the chamber, you heat it up and actually increase the pressure inside the chamber," Dr. Anand Kumar, a professor of medicine at the University of Manitoba, said. The machines heat up to about 121 C for 15 minutes, killing bacteria and viruses. "It'll sterilize anything." The assumption has been that if you tried this on an N95 mask they would degrade rapidly. We thought we'd give it a try anyway," Kumar said. "And actually what we found is while it does degrade some [types of] masks, there's a certain group of masks that are made of kind of a fabric-type material, rather than being moulded closely to the face they're called pleated [masks]," he said. Kumar said the pleated fabric masks can be cycled through an autoclaving machine 10 times and come out as good as before.

"The reason this is really important is that autoclaves are available at literally every established hospital in the world. There is probably no hospital in the world that doesn't have an autoclave machine," Kumar said. "So everybody can use this for these particular types of masks and these particular types of masks are probably the most common type of N95 mask, so we're really pleased." Kumar said the technique could be put into use at any hospital at any time. "It's a technology that's available and ready to go right now."

The new coronavirus has now infected 1 million people across the world, a milestone reached just four months after it first surfaced in the Chinese city of Wuhan. More than 51,000 have died and 208,000 recovered in what has become the biggest global public health crisis of our time. Bloomberg reports: When the virus was first discovered, doctors likened it to Severe Acute Respiratory Syndrome, or SARS, the illness that sickened 8,000 people mostly in Asia in 2003. Highly contagious, and appearing with little or no symptoms in some cases, Covid-19 has rapidly eclipsed all recent outbreaks in scale and size. Fewer than 20 countries in the world remain free of infection. With some virus carriers presenting few outward signs of illness, and many countries unable or unwilling to conduct wider testing, the true number of global infections is likely higher -- some say far higher -- than 1 million.

The U.S. now has the most cases officially recorded globally with more than 234,000, according to Johns Hopkins University, which draws on a combination of data sources -- from governments to the World Health Organization and local media -- to feed its tallies. Next is Italy, with 115,000, the JHU data show. Italy has the highest death toll with almost 14,000 virus fatalities, followed by Spain. With world travel paralyzed and millions of people under some form of lockdown as a result of government efforts to contain the spread, the health crisis has also become an economic one: The global economy is expected to shrink 2% in the first half of 2020. Business activity has ground to a halt in many sectors, with predictions the U.S. jobless rate could reach 30% in the second quarter.

AI Program Could Check Blood For Signs of Lung Cancer (theguardian.com) 19

An anonymous reader quotes a report from The Guardian: Scientists have developed an artificial intelligence program that can screen people for lung cancer by analyzing their blood for DNA mutations that drive the disease. The program works by examining free-floating DNA that circulates in the blood. The majority of this genetic detritus enters the bloodstream when harmless cells in the body break down and spill their molecular innards, but tumors also shed DNA as they form and grow larger.

Writing in the journal Nature, the scientists describe how their AI program crunched data on the DNA found in the blood of lung cancer patients to learn which common cancer mutations most effectively predicted the disease. The researchers then used the trained program to distinguish lung cancer patients from healthy people in a separate group of volunteers who gave blood samples for the study. The system cannot confidently diagnose cancer, but instead flags up likely cases for further medical investigation. In tests, the program had a 2% false positive rate -- meaning that it mistakenly flagged two in every 100 healthy people as having the disease -- while rating 55% of stage 2 cancers and nearly 70% of stage 3 cancers as patients likely to have the disease.

New X-Ray Technique Images Soft-Tissue Tumors Clearer Than MRI (newatlas.com) 16

Researchers at Tohoku University have developed a new method of adapting X-ray to image soft tissue, "so that its higher resolution can reveal tumors or other problems earlier than other techniques," reports New Atlas. From the report: Elastography is a field of medical imaging that focuses on the stiffness or softness of tissues. Shear waves are sent through the body, and then an imaging technology like ultrasound or MRI is used to watch how they spread. The waves move through stiff tissue faster than they do through soft tissue, and since tumors, lesions and hardened arteries are all stiffer than surrounding tissue, the technique can highlight these signs of disease. X-rays usually work on a different mechanism, but recent research has suggested that they could be applied to elastography too. And if they were, the resulting images would be much higher resolution, able to spot things on the scale of microns instead of millimeters.

And now, X-ray elastography has moved from principle to practice. The Tohoku team has taken the first images using the technique, and shown that it is able to identify the stiffness of different materials. The researchers imaged a polyacrylamide gel, with some samples containing harder particles of zirconium dioxide. Vibrations were then sent through these samples while X-ray images were taken. And sure enough, the X-ray elastography method was able to spot these tiny intruders. After showing that the concept does work, the researchers say that the next steps are to create 3D images, and eventually develop x-ray elastography equipment for medical diagnoses.
The research was published in the journal Applied Physics Express.

In Chicago, a nurse was fired after emailing colleagues that she wanted to wear a more protective mask while on duty. In New York, the NYU Langone Health system has warned employees they could be terminated if they talk to the media without authorization."

Doctors are a famously independent profession, where individual medical judgment on what's best for the patient is prized over administrative dictates. That's reared its head during the Covid-19 outbreak, with many physicians, nurses and other health-care workers taking to social media to express deep concerns about the lack of protective gear or much-needed patient-care equipment like respirators. Some posts have gone viral and are being shared hundreds of thousands of times, often tagged with #GetMePPE. Privacy laws prohibit disclosing specific patient information, but they don't bar discussing general working conditions.
The report notes that not all hospitals are blocking staff from talking to the press. "New York's Mount Sinai has been scheduling media interviews for nurses, physicians and trainees to help the public understand the severity of the crisis," reports Bloomberg. "The University of California San Francisco Medical Center has gotten hundreds of such calls and encouraged workers to talk to reporters."

From a report: All of that means doctors, some of whom were once reluctant to embrace social media, are wading deeper into platforms that are rife with fake news, unproven medical advice and mass panic. "Social media is the disease and the cure. It is responsible for the dissemination of misinformation as much as it needs to be a tool for repairing that," said Rick Pescatore, an emergency room physician and public health expert in the Philadelphia area, who is active on Twitter and Facebook and has treated Covid-19 patients. "It's incumbent upon physicians, who want to get real information out there, to meet these patients where they are -- and that's social media."

Perhaps nowhere is this shift more striking than on TikTok, a short-form video platform beloved by teens that is best known for lip syncing, dance routines and comedy skits. In one TikTok video viewed more than 416,000 times, a registered nurse named Miki Rai does a choreographed dance involving a lot of hand motions as facts about Covid-19 flash on the screen, such as how long the virus stays on different surfaces. In another TikTok video, set to soothing elevator music, Dr. Rose Marie Leslie demonstrates proper handwashing: Wet hands. Lather up. Start washing for 20 seconds. Scrub under your nails and between fingers. Rinse. Leslie, a resident physician specializing in family medicine at the University of Minnesota Medical School, created a TikTok account about a year ago, with the aim of reaching a younger demographic with health education information. Soon after coronavirus cases started emerging, she began creating TikToks about the issue. Now, she works to debunk myths about the virus for her more than 500,000 followers.

The esteemed science fiction author, best known for movie "Arrival" that is based on his novel, on how we may never go "back to normal" -- and why that might be a good thing. From an interview on Electric Literature: EL: Do you see aspects of science fiction (your own work or others) in the coronavirus pandemic? In how it is being handled, or how it has spread?
TC: While there has been plenty of fiction written about pandemics, I think the biggest difference between those scenarios and our reality is how poorly our government has handled it. If your goal is to dramatize the threat posed by an unknown virus, there's no advantage in depicting the officials responding as incompetent, because that minimizes the threat; it leads the reader to conclude that the virus wouldn't be dangerous if competent people were on the job. A pandemic story like that would be similar to what's known as an "idiot plot," a plot that would be resolved very quickly if your protagonist weren't an idiot. What we're living through is only partly a disaster novel; it's also -- and perhaps mostly -- a grotesque political satire.

EL: This pandemic isn't science fiction, but it does feel like a dystopia. How can we understand the coronavirus as a cautionary tale? How can we combat our own personal inclinations toward the good/evil narrative, and the subsequent expectation that everything will return to normal?
TC: We need to be specific about what we mean when we talk about things returning to normal. We all want not to be quarantined, to be able to go to work and socialize and travel. But we don't want everything to go back to business as usual, because business as usual is what led us to this crisis. COVID-19 has demonstrated how much we need federally mandated paid sick leave and universal health care, so we don't want to return to a status quo that lacks those things. The current administration's response ought to serve as a cautionary tale about the dangers of electing demagogues instead of real leaders, although there's no guarantee that voters will heed it. We're at a point where things could go in some very different ways, depending on what we learn from this experience.

China Concealed Extent of Virus Outbreak, U.S. Intelligence Says (bloomberg.com) 374

China has concealed the extent of the coronavirus outbreak in its country, under-reporting both total cases and deaths it's suffered from the disease, the U.S. intelligence community concluded in a classified report to the White House, according to three U.S. officials. From a report: The officials asked not to be identified because the report is secret and declined to detail its contents. But the thrust, they said, is that China's public reporting on cases and deaths is intentionally incomplete. Two of the officials said the report concludes that China's numbers are fake. The report was received by the White House last week, one of the officials said. The outbreak began in China's Hubei province in late 2019, but the country has publicly reported only about 82,000 cases and 3,300 deaths, according to data compiled by Johns Hopkins University. That compares to more than 189,000 cases and more than 4,000 deaths in the U.S., which has the largest publicly reported outbreak in the world.

He found himself packed into a room with about 70 other applicants, sitting shoulder-to-shoulder to watch a PowerPoint presentation about what it's like to work for the online retailer. The man, who provided a smartphone photo to document his experience, said the event was exactly like one he attended last year for a seasonal holiday job with Amazon. In other words, there were no special precautions to keep attendees safe from the coronavirus. When the man raised concerns about the crowded conditions, he said an Amazon manager mocked him and a fellow recruit sneered.

"They made jokes and told me to leave if I was unhappy," he said, adding that one manager said Amazon's operations were exempt from the rules because the company is considered an essential service. "They didn't care one tiny bit." The former customer rep took the job but still worries about getting sick. Amazon also ignored official social-distancing guidelines at hiring events near Portland, Oregon, and in Kenosha, Wisconsin, according to two applicants. A fourth person who attended an Amazon job fair in West Jefferson, Ohio, said she was sent home and asked to return another day because the gathering was too crowded, suggesting precautionary measures are in place at least at some events or Amazon is changing its practices.

The NGO Tearfund has calculated the greenhouse gas emissions from the open burning of plastic bottles, sachets and cartons produced by Coca-Cola, PepsiCo, Nestle and Unilever in developing nations, where waste can be mismanaged because people do not have access to collections. Taking a sample of six developing countries, reflecting a spread across the globe, the NGO estimated the burning of plastic packaging put on to the market by the companies creates 4.6 million tons of carbon dioxide equivalent -- equivalent to the emissions from 2 million cars.

Tearfund analyzed the plastic put on the market in China, India, the Philippines, Brazil, Mexico and Nigeria by the four companies to examine the impact of single use plastic in developing countries. The countries were chosen because they are large developing country markets, spread across three continents. The sachets, bottles, and cartons sold in these countries often end up either being burned or dumped -- creating a pollution problem equivalent to covering 83 football pitches with plastic to 10 centimeters deep each day.
The report says: "This massive plastic pollution footprint, while a crisis in and of itself, is also contributing to the climate crisis."

"These companies continue to sell billions of products in single-use bottles, sachets and packets in developing countries," says the report. "And they do this despite knowing that: waste isn't properly managed in these contexts; their packaging therefore becomes pollution; and such pollution causes serious harm to the environment and people's health. Such actions -- with such knowledge -- are morally indefensible."

Astrophysicist Gets Magnets Stuck Up Nose While Inventing Coronavirus Device (theguardian.com) 99

An Australian astrophysicist has been admitted to hospital after getting four magnets stuck up his nose in an attempt to invent a device that stops people touching their faces during the coronavirus outbreak. The Guardian reports: Dr Daniel Reardon, a research fellow at a Melbourne university, was building a necklace that sounds an alarm on facial contact, when the mishap occurred on Thursday night. The 27 year-old astrophysicist, who studies pulsars and gravitational waves, said he was trying to liven up the boredom of self-isolation with the four powerful neodymium magnets.

Reardon said he placed two magnets inside his nostrils, and two on the outside. When he removed the magnets from the outside of his nose, the two inside stuck together. Unfortunately, the researcher then attempted to use his remaining magnets to remove them. "As I was pulling downwards to try and remove the magnets, they clipped on to each other and I lost my grip. And those two magnets ended up in my left nostril while the other one was in my right. At this point I ran out of magnets." Before attending the hospital, Reardon attempted to use pliers to pull them out, but they became magnetized by the magnets inside his nose. At the hospital, a team of two doctors applied an anesthetic spray and manually removed the magnets from Reardon's nose. "Needless to say I am not going to play with the magnets any more," Reardon said.

MIT Team Shares New $500 Emergency Ventilator Design With the Public (interestingengineering.com) 107

A group of MIT scientists has created an emergency ventilator, which is affordable, and easily made using regular hospital devices. Interesting Engineering reports: A team of volunteers, scientists, physicians, and computer scientists at MIT known as E-Vent put their heads together three weeks ago to revive a 10-year-old ventilator project. The end result is a ventilator design that's affordable and easily replicated. The total cost of the device for the different parts is between $400 to $500, and the team plans on sharing their design online on their website so that manufacturers and companies can recreate the lifesaving device for hospitals around the world.

The device's main part already exists in most hospitals' inventory: Ambu resuscitation bags. Usually, these are manually operated by emergency technicians or medical professionals to keep the patient breathing until they are hooked up to a ventilator. The team at MIT has adapted the Ambu bags by attaching them to an automated mechanism that automatically pumps the bag with air in the same manner if a human were handling it. This method would alleviate the use of a person standing day and night by a patient's bedside -- something that's not currently possible in hospitals that are reaching over-capacity because of the rapidly spreading coronavirus -- and keep them breathing long enough to then be strapped to a proper ICU ventilator.
There's currently no exact date for when the prototype info will be shared for all to use, but the team members have stated that they eventually want to secure the FDA approvals.
